SIRPA

SIRPA,即信號調(diào)節(jié)蛋白α,別名Sirp-alpha-3p84、CD172a。它是一種I型跨膜蛋白,屬于免疫球蛋白超家族成員,主要在髓系細(xì)胞如巨噬細(xì)胞、單核細(xì)胞、樹突狀細(xì)胞等表面表達(dá),也在神經(jīng)細(xì)胞表面表達(dá)。SIRPA的作用機制是通過與CD47等配體結(jié)合,傳遞抑制性信號,抑制巨噬細(xì)胞的吞噬作用,從而在免疫調(diào)節(jié)中發(fā)揮重要作用。SIRPA的胞內(nèi)結(jié)構(gòu)域含有免疫受體酪氨酸抑制基序(ITIM),能夠通過去磷酸化作用調(diào)節(jié)細(xì)胞內(nèi)的信號通路。SIRPA參與了多種生理和病理過程的調(diào)節(jié),包括免疫應(yīng)答、細(xì)胞吞噬、中樞神經(jīng)系統(tǒng)發(fā)育以及定向造血干細(xì)胞移植等。SIRPA通過與CD47的相互作用,可以抑制巨噬細(xì)胞對靶細(xì)胞的吞噬,這一機制被腫瘤細(xì)胞利用來實現(xiàn)免疫逃逸。因此,SIRPA是腫瘤免疫治療中的一個潛在靶點,尤其是與PD-1-PD-L1阻斷劑聯(lián)合使用時,可能對某些類型的癌癥治療具有重要意義。

IHC image of CSB-RA443117A0HU diluted at 1:50 and staining in paraffin-embedded human stomach tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.13% DAB.

Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1μg/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG(H+L) at 1:200 dilution for 35min at 4℃.Control antibody (green line) was rabbit IgG (1μg/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
