What is TACSTD2?
TACSTD2 (tumor-associated calcium signal transduction 2) is also known as Trop-2, GA733-1 or M1S1. In addition, Trop-2 is also known as Trop-1, gp40, and KSA because of its association with epithelial cell adhesion molecules (EpCAM).
TROP2 was first identified as a membrane antigen by monoclonal antibody GA733-1, which is the origin of its name GA733-1. TACSTD2, a member of TACSTD family, is a type I transmembrane glycoprotein encoded by TACSTD2 gene. TACSTD2 represents a new cell surface receptor and plays a role in regulating the growth of tumor cells. TACSTD2 is over-expressed in many kinds of malignant tumors, and it is an oncogene related to the occurrence, invasion and metastasis of malignant tumors.
The Structure of TACSTD2
The full length of TACSTD2 gene is 9072bp, located on the short arm of chromosome 1, the specific location is 1p32.1.
TACSTD2 protein is a 36kDa polypeptide composed of 323 amino acids, which is a single transmembrane surface glycoprotein.
TACSTD2 is composed of a hydrophobic precursor peptide (AA 1-26), an extracellular domain (AA 27-274), a transmembrane domain (AA 275-297) and a cytoplasmic tail (AA 298-323).
The extracellular domain of TACSTD2 is connected to the short cytoplasmic tail by a unidirectional transmembrane helix (TM), thus the protein is immobilized on the cell membrane.
There is a phosphatidylinositol 4, 5-diphosphate (PIP2) binding sequence in the cytoplasmic tail, which is highly conserved. Besides PIP2 binding motifs, there are conserved tyrosine and serine phosphorylation sites in the cytoplasmic tail.
The Expression of TACSTD2
TACSTD2 mRNA can be detected in normal tissues, including normal keratinocytes, kidney, lung, ovary and testis. However, the expression of TACSTD2 is very low in normal tissues and overexpressed in many kinds of malignant tumors. Normally, TROP2 is mainly expressed in epithelial cells and plays an important role in the development of embryonic organs.
The Function of TACSTD2
TROP2 plays a regulatory role in cell self-renewal, proliferation and transformation. TROP2 plays an important role in embryonic development, which contributes to embryo implantation and placental tissue formation.
TROP-2 has the function of signal transduction and can be used as the transmembrane transducer of intracellular calcium signal. It is a kind of signal transduction molecule that can regulate the growth of tumor cells. It is a gene closely related to tumor. It promotes tumor cell growth, proliferation and metastasis by regulating calcium signal pathway, cyclin expression and reducing fibronectin adhesion.
TROP2 is highly expressed in reproductive system tumors, digestive system tumors, head and neck tumors and nervous system tumors, and is closely related to the occurrence of these tumors.
Applications of TACSTD2 in Treatment
As a transmembrane protein, TROP2 is overexpressed in a variety of tumors, so it can be used as a natural candidate for the development of targeted therapy. In addition, the limitation of its tissue expression reduces its therapeutic toxicity.
At present, the research targeting TROP2 has been carried out in solid tumor, gastric cancer, breast cancer, triple negative breast cancer, small cell lung cancer, non-small cell lung cancer, pancreatic cancer, urogenital system cancer, prostate cancer and other diseases.
