The DNA fragment encoding the 19-559aa of the human GRIN1 protein was fused with N-terminal 6xHis tag gene and then was inserted into the expression vector, which was subsequently transformed into the E.coli for expression. The resulting product was further purified to obtain the recombinant human GRIN1 protein. The purity of this recombinant GRIN1 protein is greater than 90% assessed by Bandscan software analysis combined with SDS-PAGE. This recombinant GRIN1 protein showed a band on the gel with a molecular weight of approximately 60 kDa.
As a novel protein of neuromuscular junction and brain, GRIN1 plays essential roles including learning, memory, and synaptic plasticity. For Alzheimer's disease (AD), GRIN1 recently has been found to play a role in regulation of AD pathway. For Parkinson's disease (PD), gene co-expression network analysis identifies that hub gene GRIN1 is related to Parkinson's disease. GRIN1 that interacts with SLC18A2 is involved in the interaction of neuroactive ligand receptors. Others suggested that GRIN1 can be associated with PD in early or later stages.

功能:Component of NMDA receptor complexes that function as heterotetrameric, ligand-gated ion channels with high calcium permeability and voltage-dependent sensitivity to magnesium. Channel activation requires binding of the neurotransmitter glutamate to the epsilon subunit, glycine binding to the zeta subunit, plus membrane depolarization to eliminate channel inhibition by Mg(2+). Sensitivity to glutamate and channel kinetics depend on the subunit composition.
