I am interested in your product ??#CSB-MP010509HU, Recombinant Human HLA class I histocompatibility antigen, alpha chain G(HLA-G) ??Could you let us know the gene introduction method to mammalian cell?
A:
Thanks for your inquiry. 1.Generally, the mammalian cells used to produce the protein is HEK 293, however, the CHO cells are also available if you have this request. 2.The transfection method used to introduce the gene into the host cell is transient transfection. 3.No virus expression system is used for this protein CSB-MP010509HU expression. 4.The commonly used vector is psectag 2A his/myc vector.
Q:
Thank you for sending the product. Unfortunately, on processing the stock we found the following discrepancy: * Product: CSB-MP010509HU (1.00mg) Recombinant Human HLA G protein (His tag) Lot number: 03853 Number of units ordered: 1 Number of units received: 1 x 1mg @ 0.15 mg/ml After centrifuging the product and then checking the content, we found that instead of the expected 6.67ml per vial, the vial received was short by 1ml. Could you please arrange for a free of charge replacement or top-up from the same lot number to compensate for this discrepancy? Many thanks in advance.
A:
Thank you for your e-mail. I checked with our tech team.? Would it be possible for you to send us a photo of your received tube? Could you please kindly check whether the tube breaks or not?? If leakage is found, we could resend extra 150ug for free to make up for this loss. If no leakage is found, because the 1mg protein was in a 10ml volume tube. Generally there is residual on the wall of tube.?Could you please check it again? We could also resend extra 150ug for free after recieving your confirmation.? Our tech team also informs that this protein was ordered frequently lately, it is out of stock. It is under production now, but lead time will be 15-20 business days.?
Non-classical major histocompatibility class Ib molecule involved in immune regulatory processes at the maternal-fetal interface. In complex with B2M/beta-2 microglobulin binds a limited repertoire of nonamer self-peptides derived from intracellular proteins including histones and ribosomal proteins. Peptide-bound HLA-G-B2M complex acts as a ligand for inhibitory/activating KIR2DL4, LILRB1 and LILRB2 receptors on uterine immune cells to promote fetal development while maintaining maternal-fetal tolerance. Upon interaction with KIR2DL4 and LILRB1 receptors on decidual NK cells, it triggers NK cell senescence-associated secretory phenotype as a molecular switch to promote vascular remodeling and fetal growth in early pregnancy. Through interaction with KIR2DL4 receptor on decidual macrophages induces proinflammatory cytokine production mainly associated with tissue remodeling. Through interaction with LILRB2 receptor triggers differentiation of type 1 regulatory T cells and myeloid-derived suppressor cells, both of which actively maintain maternal-fetal tolerance. May play a role in balancing tolerance and antiviral-immunity at maternal-fetal interface by keeping in check the effector functions of NK, CD8+ T cells and B cells. Reprograms B cells toward an immune suppressive phenotype via LILRB1. May induce immune activation/suppression via intercellular membrane transfer (trogocytosis), likely enabling interaction with KIR2DL4, which resides mostly in endosomes. Through interaction with the inhibitory receptor CD160 on endothelial cells may control angiogenesis in immune privileged sites.; Likely does not bind B2M and presents peptides. Negatively regulates NK cell- and CD8+ T cell-mediated cytotoxicity.; Likely does not bind B2M and presents peptides.
