The recombinant human Zyxin/ZYX was generated by the expression of a DNA fragment encoding 2-572AA of human ZYX in the mammalian cell. The partial-length protein carries an N-terminal 6xHis tag. It reached up to 90% in purity measured by SDS-PAGE. Under reducing conditions, a molecular weight band of about 68 kDa was presented on the gel. This protein is in-stock so that there is no waiting period for protein preparation. In addition to producing specific antibodies, this recombinant ZYX protein may be used in the studies of epigenetics and nuclear signaling.
Zyxin/ZYX is a member of the focal adhesion complex and controls cell cytoskeleton and motility. It is also involved in sensing the mechanotransduction signal. Several studies have shown that ZYX positively regulates Yki/YAP signaling. Jiuli Zhou etc. demonstrated that ZYX promotes colon cancer tumorigenesis in a mitotic-phosphorylation-dependent manner and through CDK8-mediated YAP activation. This may provide a potential cancer therapy by targeting the Zyxin–CDK8–YAP axis.
