白細(xì)胞介素
什么是白介素?
白介素,全稱為白細(xì)胞介素,是由多種細(xì)胞產(chǎn)生并作用于多種細(xì)胞的一類細(xì)胞因子。由于最初是由白細(xì)胞產(chǎn)生又在白細(xì)胞間發(fā)揮作用,所以因此得名。現(xiàn)指一類具有重要調(diào)節(jié)作用而統(tǒng)一命名的細(xì)胞因子,它和血細(xì)胞生長(zhǎng)因子同屬細(xì)胞因子。兩者相互協(xié)調(diào),相互作用,共同完成造血和免疫調(diào)節(jié)功能。白細(xì)胞介素在傳遞信息,激活與調(diào)節(jié)免疫細(xì)胞,介導(dǎo)T、B細(xì)胞活化、增殖與分化及在炎癥反應(yīng)中起重要作用。
白細(xì)胞介素interleukin 縮寫(xiě)為IL,功能關(guān)系免疫反應(yīng)的表達(dá)和調(diào)節(jié),這種調(diào)節(jié)有來(lái)源于淋巴細(xì)胞或巨噬細(xì)胞等的許多因子參與。來(lái)源于淋巴細(xì)胞的有淋巴細(xì)胞活素,來(lái)源于巨噬細(xì)胞的總稱為monokine,其中的各個(gè)因子的生物活性各有不同(例如巨噬細(xì)胞活化,促進(jìn)T細(xì)胞繁殖等),因子自身的物理化學(xué)性質(zhì)多不清楚。
白介素家族?
白介素家族成員眾多,最新數(shù)據(jù)表明,成員編號(hào)依次從IL-1~IL-38。另外,白介素受體包含三種類型,分別是I型、II型和其他類型。其中II型白介素受體包括白介素-10,20,22,28受體,其他類型包括白介素-1,8受體,而剩余的是I型白介素受體。

白介素在信息傳遞、免疫細(xì)胞激活、T細(xì)胞和B細(xì)胞活化、增殖和分化過(guò)程中發(fā)揮重要作用。這里以IL-17為例闡述IL-17作用機(jī)制。IL-17家族中有6個(gè)成員,分別是IL-17 / IL-17A、IL-17B、IL-17C、IL-17D、IL-17E / IL-25和IL-17F,由不同的細(xì)胞產(chǎn)生,主要作用是促進(jìn)促炎免疫應(yīng)答。對(duì)應(yīng)受體家族有5個(gè)成員,包括IL-17R / IL-17RA、IL-17B R / IL-17RB、IL-17RC、IL-17RD / SEF和IL-17RE。 IL-17A是該家族的標(biāo)志物,它可以保護(hù)宿主抵抗細(xì)胞外病原體,但也可以促進(jìn)自身免疫性疾病的炎癥病理,類似于IL-17C。當(dāng)IL-17F參與粘膜宿主防御時(shí),IL-17E擴(kuò)增Th2免疫應(yīng)答。IL-17家族激活MAPK,NF-κB和C / EBPs途徑中的抗細(xì)胞因子和趨化因子。Act1被認(rèn)為是該途徑中的主要介體。
