DDIT3

DDIT3,即DNA損傷誘導(dǎo)轉(zhuǎn)錄本3,別名CHOP、GADD153,是一種轉(zhuǎn)錄因子,對細胞應(yīng)激反應(yīng)尤其是內(nèi)質(zhì)網(wǎng)應(yīng)激具有關(guān)鍵調(diào)控作用。它通過與ATF4相互作用,促進自噬相關(guān)基因表達,并通過抑制mTOR信號通路激活細胞自噬。DDIT3還參與凋亡過程,通過調(diào)節(jié)Bcl-2家族和C/EBPδ誘導(dǎo)線粒體相關(guān)凋亡。DDIT3在腫瘤細胞自噬與凋亡調(diào)控中扮演重要角色,其異常表達可能增加腫瘤細胞對治療的敏感性,是腫瘤治療的潛在靶點。在谷氨酰胺饑餓條件下,DDIT3通過雙重機制平衡糖酵解和線粒體氧化磷酸化,減少活性氧產(chǎn)生,幫助腫瘤細胞適應(yīng)代謝壓力,顯示其在腫瘤細胞代謝調(diào)控中的生物學(xué)意義。

Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1μg/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG(H+L) at 1:200 dilution for 35min at 4℃.Control antibody (green line) was rabbit IgG (1μg/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
