LRRC15

LRRC15，即富含亮氨酸重復序列的蛋白15”，別名LIB。它是一種I型跨膜蛋白，屬于LRRC超家族成員之一，由人體細胞自身的基因編碼。LRRC15在多種腫瘤微環(huán)境中的基質(zhì)成纖維細胞中表達，并且其表達受TGFβ調(diào)控。LRRC15的作用機制涉及到與腫瘤微環(huán)境中的成纖維細胞相互作用，影響腫瘤細胞的遷移和侵襲，以及可能通過Wnt/β-catenin信號通路促進三陰性乳腺癌細胞的遷移和侵襲。LRRC15在生物學上的意義非常廣泛，它不僅作為腫瘤微環(huán)境中的間充質(zhì)標志物，還在腫瘤發(fā)展、轉(zhuǎn)移以及對免疫療法的反應中發(fā)揮重要作用。LRRC15的高表達與多種實體瘤類型中抗PD-L1療法反應不良相關(guān)，使其成為癌癥治療的潛在靶點。例如，ABBV-085是一種針對LRRC15的抗體-藥物偶聯(lián)物(ADC)，已在臨床前研究中顯示出對LRRC15陽性腫瘤的療效。此外，LRRC15+成纖維細胞的耗竭可以解除對腫瘤浸潤性CD8+T細胞的直接抑制，增強其效應功能，并提高對抗PDL1免疫檢查點阻斷的腫瘤消退。因此，LRRC15是腫瘤免疫微環(huán)境研究和癌癥治療中的一個重要靶點。

Immunofluorescence staining of SH-SY5Y cells with CSB-PA819484LA01HU at 1:133, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
