NT5E
CD73是由NT5E基因編碼的一種細(xì)胞表面蛋白,也是催化AMP形成胞外腺苷的主要酶。CD73廣泛表達(dá)于人體組織細(xì)胞表面,包括肝細(xì)胞、成纖維細(xì)胞、內(nèi)皮細(xì)胞、淋巴細(xì)胞和神經(jīng)膠質(zhì)細(xì)胞、近端腎小管上皮細(xì)胞等等。CD73作為代謝免疫檢查分子,可協(xié)調(diào)細(xì)胞外腺苷水平的穩(wěn)態(tài)平衡。CD73產(chǎn)生的細(xì)胞外腺苷可通過與腺苷受體(A1R、A2AR、A2BR和A3R)相互作用來激活下游G蛋白偶聯(lián)信號并調(diào)節(jié)腺苷酸環(huán)化酶(AC)活性,或者通過平衡核苷轉(zhuǎn)運蛋白(ENTs)將其轉(zhuǎn)運至細(xì)胞內(nèi)。大量研究證實,CD73在腫瘤細(xì)胞表達(dá)并釋放腺苷,以介導(dǎo)免疫逃逸,影響腫瘤發(fā)生的多個方面,例如增殖、粘附、血管生成和轉(zhuǎn)移,參與調(diào)控EGFR、β-catenin/cyclin D1、VEGF和AKT/ERK等信號通路。CD73作為一個新興的腫瘤免疫靶點,CD73抑制劑單獨或與其它抑制劑(如抗PD-1/抗PD-L1)結(jié)合,用于癌癥免疫治療已受到廣泛的關(guān)注。

IHC image of CSB-RA978310A0HU diluted at 1:100 and staining in paraffin-embedded human endometrial cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.
