TNFRSF1B

TNFRSF1B(TNF receptor superfamily member 1b)為TNF受體超家族成員,也稱為TNFR2,主要表達在腫瘤微環(huán)境中的Treg細胞和多種腫瘤細胞中(如卵巢癌和腸癌)。TNFR2與受體TNF結(jié)合后,通過激活典型和非典型的NF-kB通路來控制細胞的生存和增殖。大量研究顯示,TNFR2抑制劑能夠特異性抑制在腫瘤微環(huán)境中高表達TNFR2的Tregs的功能,同時特異性殺傷腫瘤細胞,用于抗腫瘤治療;而TNFR2激活劑則可以激活Tregs細胞,抑制效應性T細胞(Teff),進而下調(diào)免疫反應,可以用于自身免疫疾病治療。隨著對于新靶點的不斷深入挖掘,TNFR2的重要性日漸受到關。TNFR2已經(jīng)成為免疫逃逸、腫瘤生長以及檢查點阻斷抵抗性的潛在誘因。TNFR2抑制劑抗體則能夠抑制腫瘤相關Tregs的增殖和功能。
